Laryngotracheitis: reproducibility of the disease and comparison of diagnostic methods BJM

Infectious laryngotracheitis virus (ILTV) cause mild to severe respiratory disease in chickens, the purpose of our study being to use Brazilian isolate of ILTV to reproduce ILTV disease in chickens by experimental infection and to compare three diagnostic methods (nested polymerase chain reaction (PCR), virus isolation, histopathology) for detection of ILTV. Forty-eight chickens intratracheally inoculated with ILTV and a further 48 with PBS, showing mild respiratory signs 48 hours post infection (PI) but no signs of infection after day 10 PI. Every 2 days PI, six birds were arbitrarily selected from the control and infected groups, sacrificed and the trachea collected. The aim of this study was to evaluate the incidence of footpad dermatitis and quality of broilers litter fed with sorghum grain and diets based in corn. It was used 544 male and female chicks, distributed in a completely randomized design with two treatments and eight replications. The chicken feed was formulated and produced from corn, soybean meal and sorghum grain. In the formulation were kept constant levels of energy and protein in accordance with the following treatments: A. Control (diet based on corn and soybean meal); B. We have developed two loop-mediated isothermal amplification (LAMP) assays for the detection of Theileria parva, the causative agent of East Coast fever (ECF), an economically important cattle disease in eastern, central and southern Africa. These assays target the polymorphic immunodominant molecule (PIM) and p150 LAMP genes. The primer set for each gene target consists of six primers, and each set recognises eight distinct regions on the target gene to give highly specific detection of T. parva. The detection limit of each primer set is 1 fg, which is equivalent to one copy of the PIM and p150 T. parva genes. Um garanhão da raça Brasileiro de Hipismo, de 12 anos de idade, apresentou um quadro neurológico complexo, que se assemelhava, em vários aspectos, aos sinais neurológicos causados pelo herpesvírus eqüino 1 (EHV-1), variando desde uma suave ataxia à completa paralisia dos posteriores. O animal morreu com menos de 12 horas após o início dos sinais nervosos. O diagnóstico de EHV-1 foi feito pela detecção do DNA viral no sistema nervoso central por meio da reação em cadeia da polimerase (PCR) e posterior sequenciamento.

A tripanosomose causada pelo Trypanosoma evansi tem uma distribuição geográfica extremamente ampla. Ela ocorre no norte da África, Índia, Malásia, Indonésia, China, Rússia, Filipinas, América Central e América do Sul. O T. evansi causa a tripanosomose equina também conhecida no nordeste da Argentina e Pantanal como "mal de cadeiras". O T. evansi infecta uma ampla variedade de mamíferos e no Pantanal ele tem sido encontrado em cavalos, quoatis (Nasua nasua), cães, capivaras (Hydrochaeris hydrochaeris) e pequenos roedores (Oryzomys sp.). No Pantanal tripanosomose eqüina causa centenas de mortes todo ano. Esta publicação tem como objetivo descrever a metodologia do TIFI utilizada no Laboratório de Sanidade Animal da Embrapa Pantanal.

Molecular characterization of Prunus necrotic ringspot virus isolated from rose in Brazil Ciência Rural

ABSTRACT: There is no molecular characterization of Brazilian isolates of Prunus necrotic ringspot virus (PNRSV), except for those infecting peach. In this research, the causal agent of rose mosaic was determined and the movement (MP) and coat (CP) protein genes of a PNRSV isolate from rose were molecularly characterized for the first time in Brazil. The nucleotide and deduced amino acid sequences of MP and CP complete genes were aligned and compared with other isolates. Molecular analysis of the MP and CP nucleotide sequences of a Brazilian PNRSV isolate from rose and others from this same host showed highest identities of 96.7% and 98.6%, respectively, and Rose-Br isolate was classified in PV32 group.

ABSTRACT: Erwinia psidii causes bacterial blight of guava ( Psidium guajava ), an important disease of this crop in Brazil. The pathogen affects branches and twigs of guava trees, reducing yield significantly. Bacterial dissemination often occurs through contaminated but asymptomatic propagating plant material. The objectives of this research were to evaluate the use of BIO-PCR and conventional PCR to detect E. psidii in inoculated guava plants grown in a greenhouse and in symptomatic and asymptomatic trees from guava orchards. Molecular detection of protozoa of the Sarcocystidae family in sheep from the State of Rio Grande do Sul, Brazil Ciência Rural

ABSTRACT: Infections caused by protozoa belonging to the Sarcocystidae family have worldwide distribution and are common in ruminants, leading to considerable economic losses. This study evaluates Sarcocystis spp., Toxoplasma gondii and Neospora caninum infections in sheep from Southwest region of Rio Grande do Sul, Brazil. Myocardium samples of 80 sheep raised on extensive system were collected. Tissue cysts were detected by direct examination and presence of infective agents was confirmed by PCR. Macroscopic evaluation did not reveal changes, but direct microscopic examination showed cysts in 76.2% (61/80, 95% CI: 66.9 - 85.9) samples, and all cysts were morphologically similar to those caused by Sarcocystis tenella or Sarcocystis arieticanis. Aiming to improve the molecular diagnosis of symptomatic toxoplasmosis in Brazilian patients, this study evaluated the performance of real time PCR testing two primer sets (B1 and REP-529) in detecting Toxoplasma gondii DNA. ABSTRACT The fungal genus Curvularia is associated with a number of diseases in plants, commonly producing foliar spots in forage grasses. The objective of this study was to characterize the morphological and molecular diversity of the isolates of Curvularia sp. associated with Andropogon seeds, and to assess both their capacity to transmit disease and the pathogenicity of this fungus to crop. Ten isolates of Curvularia sp. were sourced from Andropogon seeds from agricultural producing regions in the Brazilian states Tocantins and Pará.
